I have added my Tax Jurisdiction/s under Tools - Tax Jurisdictions, but when I add a Taxable item to my Sales Screen, the Payment amount does not include tax.

There are two components to utilizing Tax Jurisdictions. The first one, i.e. adding the Tax Jurisdictions to Tools - Tax Jurisdictions, you have probably correctly done. The second step is to assign the apropos Tax Jurisdiction/s to the appropriate Location/s. Go Tools - Settings - Location, Modify the appropriate Location and go to the Misc Tab. Now hit the Edit Default Sales Tax Jurisdictions button and use the Arrow button to move to the right-hand screen the Tax Jurisdiction/s for this specific location.
